You want to know how the cycle was elucidated?

i may be wrong, but as far as i remember Hans Krebs and his team used some radioactive carbon to track the CO2 that came from acetyl~CoA. By bursting cells open at various times he found the Carbon atoms in various components at different times after marking. That led to finding the intermediates and the order which they are formed in. The detail were worked out later..

Im pretty sure Krebs and his team uncovered the cycle by adding intermediates of the cycle to the system and noticing how this affected the concentrations of the other intermediates.
