how to find factor which regulate target gene?

Post by thisiscq » Tue Dec 19, 2006 12:18 pm

recently, in my experiment system, we found that kinase gene Pkmyt1 is high expressed, so cell cycle is blocked at G2/M phase. my question is how to find factor which increase PKmyt1 gene expression?

There are several ways to do it... albeit all will take some time (so there is no quick/short answer).

If you think a protien is physically interacting (causing activation/inactivation) Pkmyt1 you can try to immunoprecipitate and do mass spec. (if you have access to mass spectophotometry).

You can also do a yeast 2 hybrid screen.

Anyways.. that's all that I can think of for the moment. I'll let you know if I think of anything else.. during the course of the day!

good luck!
