transcription in vitro from recombinant DNA molecules

Post by medbio » Sun Nov 05, 2006 9:38 pm

I am doing an experiment of transcription in vitro from recombinant DNA molecules.
I have to explain why transcription of uncut plasmid did not produce a clear transcript band but I have some difficulties.

I know that uncut plasmid has several distinct conformations. But is the explanation related to that? Is it because of contamination??

If someone could help me I would really appreciate it!

Thanks!
