DNA in the RT-PCR

Post by Ninking » Wed Aug 02, 2006 12:55 pm

Please help with your advice.
I can't get rid of the DNA while preparing RNA for the RT-PCR. I extract RNA with Tri-reagent (high yield). I use the DNase kit of Ambion - "DNA-free" they call it.
After the treatment I do not see DNA on a gel, but my negative control gets a band :( Negative control is a PCR with extracted and cleaned RNA without reverse transcription.
Thank you
