CO2-independent culture medium

Post by joannew » Thu Jul 27, 2006 12:15 pm

Hi, can anyone tell me what buffer is used for CO2-independent culture medium? Is it possible to use DMEM-F12 (without HEPES) as a CO2-independent medium by adding the appropriate buffer?
Thanks!

HEPES itself is a buffer. Why would you buy no HEPES and add buffer to it???

ha ha, yes I realize. But the medium I have in my fridge happens to be HEPES-free (bought for another application), just wondering if there is another buffer that might work better than HEPES.

In any case, I have tried DMEM-F12 + HEPES, and will see how the expt turned out in a few days. Fingers crossed...
