hello all
It is regarding mammalian cell growth in vitro

A cell biologist growing mammalian cells in vitro in a buffered nutrient solution
believed that
their growth was controlled in vivo by particular proteins produced by the thymus.
The experiment was planned as follows:

Thymus was dissected from the animals, homogenised in the buffer
used for growing the cells in vitro and a particular fraction obtained by
standard protein purification techniques, including precipitation, column chromatography and
fast performance liguid chromatography ()FPLC,
and ultrafiltration

A liquots of this fraction in buffer were added to the cloned cells in vitro
in this buffered solution under controlled conditions.
The same aliquots of the same buffered nutrient solution were addedd to control cells.

The dry weight of the cells was measured after 7 days.
When analysed statistically,
the mean dry weight of the cells given tha fraction was significantly greater than the mean dry
weight of the cells given only buffered nutrient solution





questions:

Q1: What could the cell biologist validly conclude from the experiment? why?

Q2: How certain is that protein(s) is/ are involved? Howcould this be tested??

Q3: What would be a more appropriate control for testing that only the thymus cell components
could have affected the cell growth??

Q4: What evidence is there that "growth" was involved in the increase in dry weight?
what would be an appropriate way to test this?

Q5: Can effects in vitro be extrapolated to effects in vivo??

note:
He should look at features of data; draw logical conclusions;
comment on the adequacy of the experiment design;
and suggest improvement in experimental design


He should examine the experimental scenario for its logic and scientific validity

The main issue is: to consider whether the cell biologist has considered the main factors affecting the system and has controlled
for them appropriately
