You asked what seems to be an easily answered question when in fact, there are so many different kinds of headaches, that the answer is
pretty complex. In general, headaches are the pain that we sense coming from pressure on nerve endings in blood vessels of the head and face, or from nerve endings in the skin and muscles of the face and skull. Tension headaches are usually due to pain from muscles whereas migraines seem to be more from blood vessels. There are also cluster headaches, hormonal and organic ones. Are they genetic? There may be a "familial tendency" for migraines but whether a gene or genes have been located, I don't know.

No direct relation, except for the one mentioned by LilKim, but you can be genetically proun to headaches if you have blood presure problems or cancer caused by genetics reasons etc.
PS: molecular genetics is nice..
