why retrovirus has to be a low copy number plasmid

Post by cwood » Fri Apr 14, 2006 4:41 pm

Recently, I start to work on Retrovirus but find the plasmid pLNCX2 vector is low copy number plasmid. Since I will use it to handle tons of different target, thought it might be convenient to make it to high copy number plasmid. Call the Clontech tech service and ask why they generate it as low copy and they told me it's happen to be the case and there is no technical reason to do so. But one of my friends told me virus vector has to be low copy due the potential self recombination rate increase if it's a high copy. Which opinion is true? Thanks for the input.
