3T3 Fibroblasts and EGF

Post by dmill4 » Thu Mar 30, 2006 2:26 pm

I'm culturing 3T3s in defined media plus EGF - I need them to proliferate in the presence of EGF and die in the absence. I grow my stock culture in DMEM with 10% FBS. But for my studies, I use a defined media plus EGF only. Over several passages, my stock 3T3s stop responding to EGF and withdraw from the cell cycle. Does anyone know why? These cells aren't suppose to change phenotypes...????

The defined media I'm using is RPMI, Serum Replacement 1 (sigma), SPIT (sigma). I'm using high EGF concentrations (20ng/mL).
