How produce seeds?

Post by HELISA » Tue Mar 21, 2006 12:58 am

Is it possible to produce seeds from mutant plants of A.thaliana that are completely sterile (they have knock-out DCL genes responsible for proper development of flowers). As far as I'm concerned it is not possible, but still I can buy seeds of these kinds of mutants. So, how they made them?

well, you can't get seeds from a sterile plant... However, If you have Heterzygous stock ... (carring a normal allele AND the mutant alle ...responsible for sterility) you would fertilize (mate) 2 heterozygous plants...

and according to a Punnet square (classical mendial genetics)

your progeny(seeds) will be:

1:4 (25%) Homozygous WT
2:4 (50%) Heterzyous (WT allele/Mutant Allele)
1:4 (25%) Homozygous mutant (Sterile)

The only caveat is that the Mutant allele (Lacking flower development) is not a dominant trait.

hope this helps!
