how is protein conformation determined with and why is it so important??

First it is determined by number,kind..sequence of amino acids that make the polypeptide chain. Polypeptide chain is folding..hydrogen bonds form between C=O and H (pleated sheet,alpha helix)..also disulfide bonds form between aminoacids that contain sulfur(cysteine). Conformation is important because function of protein depends of its shape(just look simplified key-lock model)
