I had isolated PMogen22 binary vector about 11.5 kb size and wanted to digest it with Hind III and EcorI enzymw. This is supposed to be a very simple digestion but somehow my dna gets degraded, dont know why? Have tired using kit to isolate plasmid and other protocols also but all the digestions dosent work. Can some one suggest why this happens.

Problem with your enzyme?
Have you checked mrthylation?
Hind III exhibits star activity, so do not make your digestion too long.

With any more detail I can"t say much more.
