Macromolecular Synthesis

Ich bin gerade dabei ein Protokoll zu erstellen für ein Mikroorganismus, dass nach Zugabe von Antibiotika auf DNA/RNA/Protein-Ebene mittels Tritium-markierten Metaboliten untersucht werden soll...

Meine Frage wäre, müssen sich die MO während des Einbaus von Metaboliten wie Thymidin,Uridin,Leucin in einer Wachstumsphase befinden? Also reicht es nicht wenn die MO in Phosphat-Puffer kultiviert werden und in einer Lag-Phase stehen ?

Für Ratschläge wäre ich sehr dankbar :)

so i have the question if in case of macromolecular synthesis. the cultivation of a bacterium lets say staph aureus in this experiment is denoted in many protocols in phosphate buffer...but when they do not grow in the lag phase how can tritium labeled -thymidine or uridine or leucine -be incorporate?

Wow you lot can speak different languages. Did u know that its easier to learn a new language when your like a wee bit older than a baby.
