TCR Beta Constant Regions 1/2 - Dual Expression?

Post by GavinLambert » Wed Apr 29, 2015 10:00 pm

Hi,

I wonder if anyone can help me with an incredibly frustrating issue that I simply cannot find a straight-forward answer to.

I have been working with CRISPR to knockdown endogenous T-cell receptors and have encountered decreased efficacy with my Beta guides. Now, I understand that the beta constant region contains two separate genes, TCRBC1 and TCRBC2, however I cannot see whether they are both able to be used by a single cell. Both have associated J regions, so I've been trying to explore whether there is alternative splicing or some mechanisms allowing both to be employed, which might go to explaining the reduce beta insert effectiveness.

Any links to papers MASSIVELY appreciated. I clearly no longer know how to use Pubmed...
