how to cultrue IEC-6

still now,i found that many paper mention 10%FBS+DMEM,1x105 U/liter of penicillin/streptomycin at 37°Cin
an atmosphere of 10% CO2 and 95% relative humidity. But in some forum ,someone said,1U/ml insulin must be add into the cultrue medium.
Does anyone tell me why,and what kind of medium shoud IEC-6 in?thank you

Sigma-Aldrich says: DMEM + 2mM Glutamine + 0.1 IU/ml Insulin + 5% Foetal Bovine Serum (FBS); 5% CO2

Different people use different protocols for different experiments. Sometimes they grow fine in either, but change slightly which makes one version more suitable for your experiments. If you don't know, just try different combinations and see which works best for you.

thanks for your attation,but i still don't konw why someone said insulin must add into the medium,otherwise,IEC-6 cells can not adhere to the flask.

Normally DMEM medium is modified with 4mM L-glutamin,is that necessary to add 2mM L-glutamin into medium,and what kind of insulin should i choose,you know there are lots kinds of insulin in hospital.
