Ligase reaction problem

Post by Molbio34 » Thu Feb 26, 2015 8:27 am

Dear Friends,

I uploded an image you can see below. Here, I want to ask whether it is enough base lenght for ligase reaction via thermostable ligase in 63 C (Protocol: 5 cycle at 95 C-30 s and 63 C-4 min) I think I take cross reaction because of this dimer but I am not sure. Is it possible to discard this short undesired dimers in the reaction. Thank you
