"dose"-response curves

I have always wondered why dose-response curves are constructed against the concentration of a drug rather than the amount of a drug. Typically, an effect increases or decreases with increasing concentration in g/l or mol/l. Why not graphing an effect against a mass in gram or quantity of molecules in moles ?

I have data demonstrating that increasing amount of drugs in moles exert an increased cell kill at constant drug concentration.

In terms of methodology, increasing drug amount with constant drug concentration requires to vary the volume of drug-containing medium.

I cannot explain these results. Please advise/discuss/comment!
