Need an help about Drosophila genetics

Post by golgicik » Wed Oct 30, 2013 8:46 pm

Hello!

I need help. My professor asks me this and I could not understand it. Can you please help me to figure it out?

I asked him after crossing lines with UAS-UPRT and Gal-4 driver, how are we going to select the flies? He gave me this:

Flies have 4 chromosomes: 2 and 3 are useful for genetics. For each chromosomes there are two copies just like in humans:

2nd chromosome: UAS-UPRT is homozygous

3rd Chromosome: One chromosomes carries Repo. The other carries a marker TM6 which gives flies short bristles.

You are starting from: 2nd chromosomes UAS-UPRT homozygous ; 3rd chromosomes wild-type homozygous

crossed with:

2nd chromosomes wild-type homozygous; 3rd Chromosomes Repo and TM6

Using your knowledge of meiosis, what should you be looking for?

You should be looking for heterozygot in all genes in F1.

Cis or trans? That's what matters.

Seems so to me. But they should all be the same in F1 generation.

Thank you. Actually, I am confused because of the repo and TM6 part.

It seems the answer is incorrect. I really need to figure out this. Can someone explain, please?
