Hi,

Can anyone inform me of a good webpage that describes first strand DNA synthesis? Images would be useful.

thanks

reverse transcriptase and than DNA-synthesis with a dpolyA-primer.

I think poliT primer; primer pairs with mRNA poliA and from it RT starts reverse transcription.

yes that's for the reverse transcriptase, and than i see i made a little mistake. in the process (but i was ill yesterday and it was already midnight)

But first you take a DNA-strand synthesized by reverse transcriptase. Than you digest the mRNA with alkali solution and add oligo dG at the end of the 3'-end of the cDNA.

Then you take a oligo dCprimer at the 5'-end of the 2nd strand to be produced --> result is double stranded cDNA.
