Lonza MycoAlert test

Does anyone know how robust this kit is? For example, can you answer these questions:

How many replicates of the same sample are good to test?

I've gotten false positives AND false negatives from the same sample, how to avoid false responses?

Akron Biotech makes a solution called MycoSolution3 that seems to work or at least the results comes back negative in the Lonza kit after 1 month of treatment (2 weeks on, 2 weeks off). Does the myco get completely eliminated? (It's OK if you cant answer this question)

What's your experience /frustrations with the kit?

This assay is quite robust but like all products has a few pitfalls.
1.) are you using a multifunction plate reader with a luminescence setting or a dedicated cuvette luminometer?
2.) Are you using MycoAlert or MycoAlert Plus?
Some multifunction plate readers have hopelessly weak luminescence detectors and cannot give an accurate reading, which can lead to the problems you are seeing. If you are using a multifunction, what make / model is it?
Using the MycoAlert Plus seems to help sensitivity with these weak plate readers.

As far as mycoplasma elimination goes, it is always suspect as to whether or not the infection is completely eliminated. Unless you absolutely cannot start over, I always DESTROY THE CULTURE and start over. It's the only really safe way. Lonza makes a product called MycoZap that works well at eliminating the infection without excessive toxicity to the cells in culture. Even better, they have a prophylactic version that prevents mycoplasma in the first place.
