Im trying to grow PC12 cells (from rat adrenal pheochromocytoma) on glass slides. I used another lab's protocol using poly-D lysine, washed and then added collagen. This protocol has worked for this cell line before but I cant seem to repeat it (regardless of how careful and closely I follow the protocol). I need the cells to stick for at least 10 days before I can continue with my experiment. I tried different incubation times and concentration of both poly-D lysine and collagen but i still have the same issues. Help!

How can you be sure it's not working?

Just out of curiosity since you might know this, do you know whether one actually needs to grow MDCK cells on a matrix to get them to polarize when gromw to confluency or do they secrete their own matrix (in which case one wouldnt need all that expensive coating of course). Thanx weesper
