Devising Experiments to Determine Positive Regulation

Hey members, I was having difficulity with this biology problem. I've included a picture, to avoid confusion.

For a. i would use DNA footprinting to confirm that A and B bind to the DNA whereas C would not.

However, I don't know what type of experiment I would use to determine the circumstances in B and C?

By experiment I mean, EMSA, Northern / Southern Blotting, DNase Sensitivity assay etc.

Any ideas?

you could knock-down/overexpress your proteins (A,B or C) and see, what does the level of the regulated gene X
