anyone work with SeaPlaque Agarose

Post by huyenkobe » Sat Feb 19, 2011 2:23 am

I used the SeaPlaque Agarose from Takara to do the soft agar assay. Base agar is 0.6% and top agar is 0.3%. The problem is that my gel always melted. I created base agar in the 6 wells plate and put the plate at 4oC over night. The gel solidified well when I took the plate out the 4oC, but only after 6hrs putting on 37oC all the gel melted. It so nuisance because I cannot go forward to the next top agar. How can it happen and how to solve the problem? I did it about 40 times and I was successful only 2 times. What should I do?
