1. Why does primer extension NOT work if one is trying to find the 3’ end of a mRNA transcript?

2. If you wanted to determine a transcription start site down to the very first base, when procedure would have to carry out AFTER doing primer extension?to

1. What does the Greek letter γ refer to and why is [γ-32P] ATP used?

2.In step “b”, why is [α-32P]NTP used? Specifically, what does α refer to and why is NTP rather than ATP listed?

1. think about in what direction goes the DNA Pol ;)

2. you want to determine the sequence of your mRNA. How is that process called? Read about it's limitations ;)

1. do you know, what does ATP stand for? If you don't tell us, in what process, we won't tell you (although I have a guess)

2. the same as in 1.

Cis or trans? That's what matters.
