I am in intense need of the protocol for hot start pcr, if any one knows all steps , please write here or give the reference

I think the protocol is pretty much the same as for the regular PCR. Look into the paper you got with the Pol

If you do not have a special hot start polymerase (in wax beads, attached to antibodies or any other technique) you simply add a pause after the initial denaturation at the first step of your PCR and add the polymerase manually at that moment and then start the cycling as usual. Nothing very complicated.
