GelRed distorted bands

Post by muldengold » Tue Feb 02, 2010 3:17 pm

Hi,

I was keen to try GelRed as an EtBr alternative. I simply tried some DNA ladder at various concentrations and I must say that the result was not very convincing. The sensitivity appeared to be all right, but the bands look quite distorted (like too much wet paint running down at places). I put the GelRed directly in a 2% agarose 1x TAE gel at the concentration that is recommended by Biotium (1 µl of 10,000x concentrate per 10 ml gel) and ran the gel at 100V in TAE buffer. I sent an email to the manufacturer but they haven't responded (yet). Has anyone any idea what causes these kind of warped bands and what one can do about it? Voltage too high? TAE buffer not ideal? Thanks.

Sandro
