kinase-dead Src K295M

Hi,

I have a question about a kinase-dead, dominant negative SrcK295M construct. What happens on Src phosphorylation status at Y416 (indicating active form) after introduction of SrcK295M?

Although I expected that p-Src Y416 decreases after dN-Src introduction, p-Src Y416 was increased compared to control in my expt sysem. I introduced the construct to bladder cancer cells using adenovirus vector. I wonder if SrcK295M products might be phosphorylated at Y416 but I'm not sure. Could anybody know about this?
