Competent cell preparation

Post by jshreyas » Thu Jan 14, 2010 6:36 pm

I have prepared SOB for competent cells preparation. According to the procedure, I have to add Mg solution (MgSo4.7H2O and MgCl2.6H2O) after autoclaving it (just before use). but by mistake, I added Mg solution before autoclaving it. I can always discard it and make a fresh SOB solution, but is there any other way through I could save the reagent from getting wasted. What would happen to divalent ions if I add them and then autoclave the solution?

They will lose the water  (that said one postdoc in our lab)

IMHO nothing.

BTW what's the difference between SOB and SOC?

Cis or trans? That's what matters.
