hygromycin as selection marker

hi

I transformed my plant callus with a gene construct that included hygromycin resistance gene... grew my plants on hygromycin containing media and now did a pcr for the hygromycin gene ( isolated dna from the seedlings that grew on the media)... but no hygromycin resistance gene is there on the gel...

how can it be that even though the plants dont contain hygromycin resistance gene they are still able to grow on the media?

is there a disadvantages to using to much hygromycin...can it harm cells that have resistance gene?
