How to extract cellulomonas bacteria and rid the fungi?

Post by Panen » Wed Nov 21, 2007 2:08 pm

I'm doing a school project. I'm trying to extract cellulomonas bacteria from compost material, but all that keeps appearing in our medium is lots, and lots of fungi. We have tried enrichment, yet the fungi keeps appearing and we can't isolate the bacteria. Any tips?

Well, just a quick suggestion: make several dilutions of your material and spread them on the growth agar or whatever you use (I suspect you use/can use solid medium - use the standard spread culture method). At least on a very diluted sample, or after several dispersion steps, you should be able to see colonies emerging from single cells. Just pick a couple of these clonal colonies, plate them, and see how they look. You should be able to exclude any fungi relatively easily this way, even if it was very dominant in the starting material.

If you have no luck with this, then I have to think this further :-P

if you are really desperate i believe there are some chemicals that specifically inhibit EK ribosomes, so spreading some of that on your plate will kill all the fungi. Don't remember exactly what the names are, but i think you can find them pretty easily. Buying them would probably be much more complicated.
