Hi guys! I'm trying to transfect HeLa cells with STAT1-conjugated ECFP. I use a plate of about 2 square cm, transfecting with 500ng of DNA and 2uL of lipofectamine LTX per transfection. It seems that I'm getting transfection, as I can see ECFP under the microscope, but the cells seem apoptotic (weird nucleus, shape is changed...). I used to do the transfection overnight, so about 20-24 hours, but have found that doing it 6hours i get as good transfection efficiency, but the cells are not as dead. Even with the 6 hour transfection period, the cells are apoptotic. Any ideas? Thanks!
