A question about cytokine production, please help!

My macrophage cell line (mouse J774) used to produce a high amout of TNF-alfa upon LPS stimulation (last Nov). But now their response is so weak (50 fold lower)! I even thawed the earliest stock I have yet it didn't change much. Does anyone know whether it's likely that cell lines could readily change their cytokine production? Anything I should pay particular attention when culturing them in order not to alter its immunopotency? I checked the cell morphology and they look normal, w/o apparent contamination.

are the cells still healthy? you are getting a low concentration of cytokines right? How is the culturing going
