DNA concentration measures

Post by Susana » Tue Apr 24, 2007 1:33 pm

Hi,
I work with plants and I use a CTAB protocol to extract DNA. When I determine DNA concentration by comparision with lambda DNA the value is significantely inferiour to that obtained in a nanodrop. Why? What kind of contamination I have in my simples?
Thanks
Susana

Depends on your OD 260/280 and OD 260/230 (for protein and carbohydrate contamination if I'm not mistaken) but look this up in any good lab manual like maneatis
