MJ PCR machine PT200 gradient question

Hi there,
I'm working in a new lab and they have the MJ research PT200 thermocycler. The dual block holds 48 0.2 tubes on each side, 6 across and 8 down. Does anyone know the orientation of the block so I can set up a gradient? The manual doesn't show the dual 48 well block.

Hi jamiejlg,

We have two of the same machines. I may be wrong, but you may not be able to do gradients with the two 48-well block on the machine. When we had only one machine, we had two separate blocks, one with the two sets of 48 wells, where you can run two different programmes at the same time, and a second 96-well block for doing gradients. We had to switch these "alpha blocks" to do normal PCRs and gradients. The manual must say something about this.

Hi- thanks for your reply. I contacted a rep- the machine I have is a gradient, but the manual just shows the gradient along the 12 well side. According to the rep- for the split block, the gradient runs along the 8 well side. Hopefully, anyway- since that's what I'm going to try!

That's good to know. Did the rep say if you could do two gradients at the same time on the split block? That would be very convenient for us.
