Plasmids for beginners...?

Post by Ammie » Fri Mar 16, 2007 4:47 pm

Do you know if there is any good "which plasmid should I choose"-guide for beginners? Don't know that much about plasmids and how to think.

I want to clone a ca 7000bp sequence. (Large sequence - low copy plasmid?).

And I want a suitable promotor. (How do I know which is suitable?)

And what more to think about except that it would be good to have restriction sites that differs on the two sides. (Guess I will have to clone it in two steps because of it's lengths?)

thanks

Check Promega site they have a large list of plasmids and plasmid info. Invitrogen may also be a good starting point.
7,000 is not too large you should not have any major issue in cloning this
pBluescript is a widely used plasmid
