Hi,

I am facing some problems when I use HIS3-MX6 selection for budding yeast transformations. I was wondering if anybody had similar problems and would have a solution (like a specific way to transform yeasts with plasmids containing HIS3-MX).
I try to transform an integrating PCR fragment that contains HIS3-MX6 marker but I don't recover any transformants on -HIS drop-out plates. (I do not have any problems for my other transformations).
