stem cell identity.

hi all i have searched all websites and textbooks but cannot find a good answer to this question. it is describe how genetic methods were used to identify and investigate gene controlliing stem cell identity? the only one i found out was pokymerase chain reaction(pcr). any help please??

You should have studied in your course something related to this question, you should have some direction. You should start thinking about a fool experiment and start describing it.
I'll start:
Take stem cells; put them in a medium which will cause differentiation (for example muscle cell extract to induce differentiation to muscle cells). Now you have two populations differentiated and non differentiated cells.
What's next?
You said PCR, how would you employ the technique, which primers will you use on which cells?
Maybe there's something else?
