Genetic mapping

Hey guys. I need to know how one can locate a mutation that can lead to a disease. Is it by positional cloning? (only genetic mapping, no physical mapping)

yes.. you could find a location by positional cloning

Are there any other methods? Or is positional cloning the main method? I believe there are several subsmethods under positional cloning?
