Plant will get cancer by infection or injure. But it is different from human cancer. It dose not spread to other position of the plant.

It is depend on what you want to do in future. If you want do research in labs, you pay more attention on your interest. If you only want to learn something and find another jobs in company, I think, prestigious field or bio-medicine would be better choice.

Don't choose high concentration of imidazole, just at the concentration you could wash the protein down. High temp. could also lead to activity losing, so keep the process at low temp.

For step 2, you could change the temp. gradient according to your sequences properties. lower of higher.
