merv merv merv! Thanks alot for the comprehensive troubleshooting notes. Yes you are right, I didnt design the primers however I tried the same experiment with a different gene of interest and I got the control result I was looking for. I am using actin for plant cell genome as its the most favorab...

Re: general QPCR problem

How are you making your message, as the other commentator wrote, you may be amplifying from genomic DNA (gDNA). This is one of the reasons for the actin control, so it is vital that you get a sample free of this if this is a problem. That's not true. The actin is used to compare different levels of...

Re: Issues of EST in whole genome gene discovery

However, there are limitations of this technology. Of course the advantage of EST is that it provides direct evidence of the expression of a gene. I understand what you are saying, but most people are only interested in whether you get an active protein or not - you say expression of a gene - but r...

Re: Re:

Ok, so this breeding program of yours. Have you considered invading Poland first? All in due time, my friend. First we need to have finished breeding our army of gargantuan Southern Elephant Seals before any invading can be done. After that we will repopulate the country with very tall and beautifu...

Re:

this article is stupid. It claims that when a gene is duplicated, both copies are ALWAYS under purifying selection. That cannot be true. If it were, then: 1. pseudogenes wouldn't exist 2. down syndrome wouldn't be bad - duuuude....what the f are you saying? next you'll be saying having slanty eyes ...

Let me simplify it for you - all of life needs a niche. Humans were driven out of the trees due to their ineptitude there. They were unable to compete. Humans thus prefer trees to be chopped down due to the D-EEEP psychological scars. The rest of the animals appreciated that they were a great food s...

A lot of the problems as I see it relate to humankinds obsession with its own abilities - like Narcissus, it has fallen in love with its own beauty, intellectually speaking. In doing so, it has lost touch with alternative viewpoints, no more evident than in religious dogma; ironically, only in its c...
