Recommendations for Bacteria Detection?

... present in the water. But it is difficult to count the motile bacteria hemocytometer. I suggest to use a disinfectant to kill the bacteria before counting.

PFGE with Haemophilus influenzae

I am doing PFGE with Haemophilus influenzae. Also i am using SmaI restriction enzyme, but i can not get result. Not as smear, actually i am counting 6 bands but as we know accoding to Tenover criteria we have to find 7 bands. I mean i have to find minumum 8 bands. Any idea about band profile ?
