thank you mr jack bean. i'm thinking about using pRSET vector for overexpression. and spread plate of LB + amp to select the transformants. i think all metods step by step i'd get from my research in the net and from other sources. one thing that give me a headache is how i have to do ...

Help Needed - Ligation Pairing (Vector and DNA)

... general, which of the above possibilities would be the best ap- proach in a subcloning experiment like the one you have done? Why? 2. Assume the transformants produced with the ligated DNA were also plated on ampicillin medium. Would you expect to see a significant difference in the number of ...

Problem with SmaI restriction digest

... band, in contrast to the double band of the undigested vector, but if I transform the digested, unligated vector into E. coli I get heaps of transformant colonies, which seems to indicate that there is a very high background of uncut vector. Can anyone help me work out what the problem is ...

help:double enzyme digests,ligation,but transformant wrong.w

NdeI and SacII double digests and then divided my vector and segment(both 3kb),ligation at 4degree. finally, got several transformants(6kb).select two enzymes(both vector and segment have their sites) cut these transformants, only got 6kb segment, no small segment.I selected other two enzymes to check...
