Thrombin

thrombin

(Science: enzyme) protease (34 kD) generated in blood clotting that acts on fibrinogen to produce fibrin. Consists of two chains, A and B, linked by a disulphide bond. B chain has sequence homology with pancreatic serine proteases: cleaves at arg gly.

Thrombin is produced from prothrombin by the action either of the extrinsic system (tissue factor _ phospholipid) or, more importantly, the intrinsic system (contact of blood with a foreign surface or connective tissue). Both extrinsic and intrinsic systems activate plasma factor X to form factor Xa which then, in conjunction with phospholipid (tissue derived or platelet factor 3) and factor V, catalyses the conversion.

thrombin cleavage

A protein of interest is bound to a GST tag. I want to separate the GST tag by using a protease. I found out that thrombin has two cleavage sites in the protein sequence whereas trypsin had more than 20 cleavage sites. I should pick thrombin over trypsin because it is more specific. ...

Struggling with Vector Cloning Project

... I don't know how far you need to take it, but if you were interested in purifying FAS2, you would have to take the fusion protein and clip it with thrombin and then re-purify the FAS2 (or purify away the GST, whichever is easier).

ENZYME CASCADE

... in great detail. A very simple explanation we had in our bio lecture was that cells at the site of injury release clotting factors that convert prothrombin to thrombin, which in turn converts fibrinogen to fibrin, which helps platelets to form a clot.
