Re: Isolation of Mitochondria... should have pellet rich in mithocondria (and chloroplasts if you are talking about plant cells.) Then, extract the mithochondria carefully with a syringe or a vacuum-pipe, and you should have the mithocondria pretty isolated.

Mouth or Nose?... showed that the outcome was the same; regardless of suctioning. To aswer your question, the mouth is always suctioned first 3-4 times with a bulb syringe. Can you reuse qiashredder columns?... they shouldn't bind nucleic acid. I don't see why you can't just was them really well, or just pull your sample through a 22guage or smaller syringe.
