... got double band align under each other in Agarose Gel when I load DNA PCR Product ??? Knowing that this DNA is Human DNA and I got it from Buccel swab and I am working in SNPs sequencing. Thanks in Advance.

if you want to use known bacteria then get your hands on some ecoli and swab it on an agar plate or just wipe your arm pits with a cotton bud and wipe it on the agar plate you'll be suprised what turns up. :D :D :) :) :mrgreen:

You don't typically Gram stain a throat swab (whatever wikipedia says). That is a pretty useless effort due to the variety of bacteria that can be observed the inability to differentiate esp among the Gram positive cocci. You would streak for ...

I need help determining what bacteria I have found on agar

And I have no clue how you come to the conclusion of a protist (fungi/mold are also quite likely on this kind of swab), neither do I see what a fuzzy foot can be when it refers to anything else than a hobbit.
