Results from our forumgrowing E coli from a starter culture... can go directly from glycerol to broth. -but if you have enough time streak on a plate and restart from an isolated colony. and ampicillin stinks ... try to go with a first isolation/selection step before restarting a culture.

Streaking for Isolation... want is to significantly decrease the number of bacteria between each streak (or set of streaks). So the simple reliable and quick method I used ... across the reamining space. I could usually isolate easily 6 different culture on 1 plate with a good reliability.
