rest

1. To cease from action or motion, especially from action which has caused weariness; to desist from labour or exertion. God . . . Rested on the seventh day from all his work which he had made. (gen. Ii. 2) Six days thou shalt do thy work, and on the seventh day thou shalt rest. (Ex. Xxiii. 12)

Annealing Temparature

The primers obviously contain added restriction sites and the matching part is quite short. I would anneal the first cycle at 50°C and the rest at 65°C. Denaturation for 10s will be enough, but a bit higher temperature 95-98°C might be better. ...

Increase in free energy in glycolysis

... biology textbook. It says that in the first 5 steps the free energy increases with each step. Two of those steps are powered by ATP, but the rest are not. In those other steps, how does the free energy increase? What powers it? Thanks.
