Do we know all the proteins (and their primary structure!)?

Why should it be impossible? Even nowaydays we are often able to predict the splicing. And posttranslational modifications are not part of primary structure (unless it's Hyp in collagen as mentioned above)

Need help with translation of DNA

... of the gene, it is just Met. Usually the first START codon is true START codon (but codin for Met anyway) and unless he didn't provide data about posttranslational modifications, you should leave it there. The same with STOP codon. The first works as STOP codon and the others are just for case... ...

When are parts of prokaryotic operons seperated?

... gene sequence in the mRNA, so that the ribosome can bind on more than 1 place on the mRNA? Or is it translated also as 1 unit and then cut of in posttranslational processing? / Thanks
