Phase plate

phase plate

(Science: microscopy) The plate used near the back focal plane of a microscope objective lens (in conjunction with an annulus at the front focal plane of the condenser lens) to achieve phase contrast. The phase plate selectively shifts the phase of the waves diffracted by the specimen by a quarter wave and reduces the amplitude of the undeviated, direct beam.

Question about chromosome replication before mitosis

... the answer lies in the much historical definition of a chromosome. In S phase, each chromosome makes a copy of itself, but the two copies (called ... level. Now, in metaphase the chromosomes line up at the metaphase plate. At precisely the same time they split into the two daughter chromosomes ...

How to practically carry out Bacterial Transformation

... For this we have to treat the grown bacterial cultures at its log phase of growth, with CaCl2. Centrifuge the cells growing at the log phase ... be transformed by intercalating with its genome. These cultures can be plated on a selection agar plate and the transformed colonies can be selected ...

How do you grow an isolated colony of e.coli from scratch?

... is great but expensive I usually isolate 6 different colonies per plate :) and in BHI or LB 16h is more than enough to get nice E. coli colonies. ... survival rate if you use a colony in exponential or late exponential phase (OD~0.6) but you will have more than enough with a saturated culture. ...
