... it's a risky method to control baby, but for your information, I don't think you can get pregnant as his sperms has been exposed to air for such a long time.

... that dense in terms of colony numbers. The plates are only grown overnight at 37 degrees so I don't think its that they are being incubated too long. What might be the cause of this? The only thing I can think might be the problem is her antibiotic being degraded/the wrong concentration? (ampicillin) ...

MOLECULAR CLONING

The purpose of dephosphorylation is to prevent self-ligation of empty vector. So unless your insert is very long, it should not be necessary.
